John Sherman Cooper (23 Agustus 1901 – 21 Februari 1991) adalah seorang politikus, ahli hukum, dan diplomat Amerika Serikat dari Persemakmuran Kentucky. Ia menjalani tiga masa jabatan parsial dan tidak berturut-turut di Senat Amerika Serikat sebelum terpilih untuk dua masa jabatan penuh pada tahun 1960 dan 1966. Ia juga menjabat sebagai Duta Besar AS untuk India dari tahun 1955 hingga 1956 dan Duta Besar AS untuk Jerman Timur dari tahun 1974 hingga 1976. Ia adalah orang Republik pertama yang terpilih secara populer untuk lebih dari satu masa jabatan sebagai senator dari Kentucky dan, pada tahun 1960 dan 1966, ia mencetak rekor margin kemenangan terbesar bagi calon senator Kentucky dari salah satu partai.
